摘 要:
以烟草原生质体为材料, 采用彗星电泳检测用 0.5 W•m-2 紫外线以不同时间(0、5、10、30、60 和120 s)诱导的烟草原 生质体中 DNA 的损伤。结果表明, 在 0~10 s 的时间内代表 DNA 损伤程度的尾矩、Olive 尾矩等参数与紫外线照射时间具 有良好的时间依赖关系。本文建立的烟草原生质体体系采用彗星电泳技术, 可以快速而灵敏地检测紫外线对植物细胞的损 伤程度。关键词:彗星电泳; DNA 损伤; UV-B 辐射; 烟草原生质体

Abstract:
Protoplasts of Nicotiana sylvestris was used for experimental material to investigate DNA damage in protoplast which was induced by 0.5 W·cm-2 UV-B for different time (0, 5, 10, 30, 60, and 120 s) by using comet assay. The results showed that the indexes of tail moment and olive tail moment etc which detected the DNA damage had statistically dose effect relationships with the time of UV-B radiation from 0 to 10 s. Comet assay detected the degree of DNA damage of tobacco protoplast induced by UV-B radiation rapidly and sensitively.Key words: comet assay; DNA damage; UV-B radiation; tobacco protoplast
